dc.description.abstract | Epilepsy is a complex and multifactorial neurodegenerative disease of which the most prominent feature is the recurrence of seizures. Oxidative stress plays a vital role in the induction of epilepsy. Daidzin is well-known for its effective ant-inflammatory and anti oxidant potential from centuries. Therefore, current Shldy focused on exploring the anti epileptic potential of daidzin in PTZ induced mice model by mainly targeting on oxidative stress. Daidzin (I , 5 and 10 mg/kg) was administered for acute study and dose was optimized. Pretreatment with daidzin remarkably reduced the score of epileptogenesis in a dose dependent manner. Chronic epilepsy was developed in mice by administration of PTZ (35 mg/kg, i.p) every alternative day for twenty-one days. Daidzin significantly prevented epileptogenesis and reversed histopathological changes associated with oxidative stress. Moreover, daidzin remarkably improved the brain histopathology by enhanced antioxidant (glutathione, glutathione sulfo-transferase, superoxide dismutase and catalase) levels. Furthermore, daidzin significantly decreased production of stress inducible enzyme and nitrite level in brain. Additionally, daidzin remarkably reduced the frequency of seizures and improved the latency to the beginning of seizure. The oxidative stress was remarkably reduced by decrease in lipid peroxidation level assessed by MDA assay. Spectroscopic analysis (FTIR, and DSC) were also performed to evaluate the efficacy of compound. Inclusively, the current data demonstrated that daidzin has remarkable neuroprotective and anti-epileptic properties through modulation of oxidative stress. | en_US |
